Mechanism and Biochemical Profile

Ganetespib binds to the N-terminal ATP-binding pocket of heat shock protein 90 (HSP90), thereby destabilizing and promoting the proteasomal degradation of key oncogenic client proteins—including HER2, mutated EGFR, Akt, and c-Met—which disrupts downstream pro-survival signaling pathways and triggers apoptosis in cancer cells. This molecular degradation of multiple driver kinases provides the mechanistic foundation for inhibiting tumor proliferation in clinical trial settings such as breast cancer, small cell lung cancer, and sarcomas.
