Biological Activity

Ganetespib (STA-9090) is a potent, triazolone-containing heat shock protein 90 (Hsp90) inhibitor that binds to the N-terminal ATP-binding domain of Hsp90. By inactivating Hsp90, ganetespib promotes the rapid degradation of multiple oncogenic client proteins, including EGFR, HER2, Akt, and MET. This loss of client protein signaling disrupts downstream oncogenic pathways, resulting in cell cycle arrest and induction of apoptosis across diverse cancer cell models. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
