Biological Activity

(E/Z)-BCI is a cell-permeable allosteric inhibitor of dual-specificity phosphatase 1 (DUSP1) and dual-specificity phosphatase 6 (DUSP6). By blocking phosphatase activity, (E/Z)-BCI hyperactivates ERK signaling downstream of the MAPK pathway. In H1299 human lung cancer cells, this compound induces cell death and apoptosis through the enhanced generation of intracellular reactive oxygen species (ROS) and activation of the intrinsic mitochondrial pathway. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
