Dinaciclib (SCH 727965) is a potent inhibitor of CDK2 (IC50 1 nM), CDK5 (IC50 1 nM), CDK1 (IC50 3 nM), and CDK9 (IC50 4 nM). It blocks thymidine incorporation into DNA and suppresses retinoblastoma protein phosphorylation. By disrupting cell cycle progression and RNA polymerase II transcription, Dinaciclib induces apoptosis through caspase-8 and caspase-9 activation across a broad spectrum of human tumor cells. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
