Biological Activity

Daraxonrasib (RMC-6236) is a RAS(ON) multi-selective, noncovalent tri-complex inhibitor targeting the active, GTP-bound state of mutant and wild-type canonical RAS isoforms. By binding cyclophilin A to form a ternary complex with RAS(ON), it sterically blocks effector engagement and inhibits downstream RAS/MAPK pathway signaling. Consequently, this targeted inhibition suppresses cell proliferation and induces marked tumor regression in RAS-addicted cancer models, particularly those with KRAS codon 12 mutations. [1][2]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
