Biological Activity

Calcium folinate (Leucovorin calcium) is a reduced folate derivative and metabolically active coenzyme precursor that bypasses dihydrofolate reductase inhibition. In folate pathway biochemistry, it expands the intracellular 5,10-methylenetetrahydrofolate pool, which stabilizes the ternary complex formed with fluorodeoxyuridylate and thymidylate synthase. This sustained interaction enhances thymidylate synthase inhibition, suppresses DNA synthesis, and potentiates fluoropyrimidine-mediated cytotoxicity in cancer cells. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
