AG-120 (Ivosidenib) is an orally bioavailable, small-molecule inhibitor of mutant isocitrate dehydrogenase 1 (IDH1). By selectively inhibiting mutant IDH1, AG-120 blocks the reduction of alpha-ketoglutarate to the oncometabolite D-2-hydroxyglutarate (2-HG). This reduction in cellular 2-HG levels promotes demethylation of histones and DNA, reversing malignant epigenetic reprogramming and inducing cellular differentiation to suppress tumor growth. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
