Biological Activity

ZZW-115 is a potent nuclear protein 1 (NUPR1) inhibitor. By binding to NUPR1, ZZW-115 competes with importins to prevent NUPR1 nuclear translocation, thereby disrupting downstream stress response networks. This inhibition causes mitochondrial dysfunction, ATP depletion, and reactive oxygen species accumulation, culminating in cell death via necroptosis and apoptosis as well as robust antitumor activity in vivo. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
