Zotatifin (eFT226) is a sequence-selective inhibitor of the RNA helicase eIF4A. It promotes eIF4A binding to specific mRNA sequence recognition motifs within 5'-untranslated regions, disrupting the assembly of the eIF4F initiation complex. By promoting this interaction, zotatifin acts as a sequence-specific translation repressor that downregulates key oncogenic driver proteins and demonstrates potent anti-tumor activity. [1]

A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
