Biological Activity

YDR1 is a selective, orally bioavailable Cereblon-based proteolysis targeting chimera that acts as a potent degrader of SMARCA2. By targeting SMARCA2 for proteasomal degradation, this compound disrupts SWI/SNF chromatin remodeling complex activity, downstream gene expression, and oncogenic signaling pathways. Consequently, SMARCA2 degradation exhibits robust antitumor activity and induces synthetic lethality in SMARCA4-mutant non-small cell lung cancer xenograft models. [1][2]
