Biological Activity

Y-39983 Dihydrochloride is a selective Rho-associated protein kinase (ROCK) inhibitor with an IC50 of 3.6 nM. By suppressing ROCK signalling, it regulates actin cytoskeleton organization, cell contraction, and neurite outgrowth. In ocular tissues, inhibition of the ROCK pathway by Y-39983 promotes trabecular meshwork relaxation, enhances aqueous humor outflow to lower intraocular pressure, and stimulates axon regeneration and survival of retinal ganglion cells. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
