WNK463 is a potent pan-WNK kinase inhibitor that targets WNK1 (IC50 5 nM), WNK2 (IC50 1 nM), WNK3 (IC50 6 nM), and WNK4 (IC50 9 nM). By inhibiting WNK family kinases, it disrupts the downstream OSR1/SPAK signaling pathway, which regulates ion cotransporters in the kidney. This targeted inhibition modulates renal sodium reabsorption and potassium excretion phenotypes. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
