Biological Activity

WH-4-023 is a potent inhibitor of Lck (IC50 2 nM) and Src (IC50 6 nM). It also potently inhibits SIK1 (IC50 10 nM), SIK2 (IC50 22 nM), and SIK3 (IC50 60 nM), displaying over 300-fold selectivity against p38α and KDR. By blocking SIK signaling, WH-4-023 suppresses CRTC3 phosphorylation, promoting nuclear translocation and enhancing anti-inflammatory IL-10 expression in macrophages. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
