Biological Activity

VU0463271 is a potent antagonist of neuronal-specific potassium-chloride cotransporter 2 (KCC2) with an IC50 of 61 nM, demonstrating over 100-fold selectivity over Na-K-2Cl cotransporter 1 (NKCC1). Blockade of KCC2-mediated chloride extrusion disrupts intracellular chloride homeostasis and impairs GABAergic inhibitory neurotransmission. This disinhibition causes a depolarizing shift in GABA reversal potential, leading to enhanced neuronal spiking and network hyperexcitability. [1][2]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
