VU-29 is a selective positive allosteric modulator (PAM) of the metabotropic glutamate receptor 5 (mGlu5). By enhancing mGluR5 receptor activity in response to glutamate, this compound modulates downstream intracellular signaling pathways, including phospholipase C and protein kinase C activation. Potentiation of mGluR5 signaling regulates excitatory synaptic transmission, neuronal excitability, and synaptic plasticity across various central nervous system pathways, serving as a valuable tool for investigating mGlu5-mediated neurotransmission and neuropharmacology. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
