VTP50469 is an orally active, highly selective inhibitor of the Menin-MLL interaction (Ki 104 pM). By targeting menin, it disrupts the Menin-MLL complex and alters chromatin accessibility, leading to downregulation of oncogenic driver genes such as HOXA9 and MEIS1. This inhibition suppresses leukemic cell proliferation, induces cell differentiation, and demonstrates marked anti-leukemia activity in MLL-rearranged leukemias. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
