Vidofludimus (4SC-101) is an orally active, potent dihydroorotate dehydrogenase (DHODH) inhibitor that disrupts de novo pyrimidine biosynthesis. By selectively inhibiting human DHODH activity, it suppresses activated lymphocyte proliferation and reduces the expression of pro-inflammatory cytokines, including IL-17A, IL-17F, and IFN-γ. Consequently, vidofludimus attenuates inflammatory signaling pathways and suppresses colonic inflammation in animal models of inflammatory bowel disease.
