Biological Activity

Varenicline (CP-526555) is a highly selective nicotinic acetylcholine receptor partial agonist targeting α4β2 receptors, exhibiting high selectivity over α3β4, α7, and muscle-type nAChRs, while also acting as a 5-HT3 receptor agonist. By partially activating α4β2 nAChRs, varenicline modulates cholinergic signaling and stimulates mesolimbic dopamine release, while simultaneously blocking full agonist binding to diminish nicotine-induced reward phenotypes. FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
