Biological Activity
UNC669 is a potent and selective malignant brain tumor (MBT) domain inhibitor targeting L3MBTL1 with an IC50 of 6 μM, displaying 5-fold selectivity over L3MBTL3 and 11-fold selectivity over L3MBTL4. By selectively binding to the MBT domain of L3MBTL1, UNC669 disrupts the recognition of methylated lysine residues on histones, leading to altered chromatin structure and modulating downstream transcriptional repression. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
