ULK-101 is a potent and selective autophagy inhibitor targeting ULK1 (IC50 = 8.3 nM) and ULK2 (IC50 = 30 nM). By suppressing ULK1/2 kinase activity, ULK-101 blocks the initiation of autophagy and inhibits downstream autophagic flux in cancer cells. Inhibition of this pathway sensitizes cancer cells to nutrient starvation and cytotoxic agents, compromising cellular survival under metabolic stress.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
