Biological Activity

Trimethoprim is a synthetic bacteriostatic antibiotic and potent inhibitor of bacterial dihydrofolate reductase (DHFR). By selectively binding to bacterial DHFR, it halts the reduction of dihydrofolic acid to tetrahydrofolic acid, thereby depleting necessary precursors for bacterial thymidine, purine, and DNA synthesis. This disruption of the folate biosynthesis pathway leads to the inhibition of bacterial cell growth and division across various Gram-positive and Gram-negative pathogens. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
