Mechanism and Biochemical Profile

Triapine potently inhibits ribonucleotide reductase (RNR) by chelating essential iron ions at its active site, thereby preventing the conversion of ribonucleotides to deoxyribonucleotides and halting dNTP pool synthesis required for cellular DNA replication and repair. This interruption of genomic duplication causes cell cycle arrest and triggers apoptotic pathways in rapidly dividing cells, providing a targeted mechanism for inhibiting tumor growth in clinical contexts such as advanced pancreatic adenocarcinoma and various hematologic malignancies.
