Biological Activity
Trastuzumab emtansine (T-DM1) is an antibody-drug conjugate (ADC) comprising the HER2-targeted monoclonal antibody trastuzumab linked to the cytotoxic agent DM1. Upon binding to HER2 on tumor cells, T-DM1 undergoes receptor-mediated internalization and lysosomal degradation, releasing DM1 catabolites that disrupt microtubule polymerization. Additionally, T-DM1 retains trastuzumab-mediated inhibition of HER2 signaling pathways, such as AKT/mTOR, and induces antibody-dependent cell-mediated cytotoxicity, ultimately leading to mitotic arrest and apoptosis.[1][2]
