Biological Activity

TOFA is an allosteric inhibitor of acetyl-CoA carboxylase-α (ACCA). Intracellularly converted to TOFyl-CoA (5-tetradecyloxy-2-furoyl-CoA), TOFA exerts potent allosteric inhibition on ACCA to effectively block downstream lipid biosynthesis pathways. This blockade suppresses de novo fatty acid synthesis, resulting in cell cycle arrest, impaired cellular proliferation, and the induction of apoptosis in a dose-dependent manner. [1][2]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
