Biological Activity
Tiamulin is a semisynthetic pleuromutilin antibiotic that target-selectively binds to the peptidyl transferase center on the 50S ribosomal subunit. By interfering with acceptor and donor substrate positioning within the ribosomal peptidyl transferase center, it selectively blocks peptide bond formation during translation. This inhibition halts bacterial protein synthesis, leading to potent antibacterial activity against susceptible Gram-positive bacteria, mycoplasmas, and fastidious animal pathogens. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
