Thiomyristoyl is a potent, selective SIRT2 inhibitor (IC50 28 nM) that also inhibits SIRT1 (IC50 98 μM) while showing no inhibition against SIRT3 at 200 μM. By targeting SIRT2, Thiomyristoyl disrupts sirtuin-mediated deacetylase activity, leading to increased lysine acylation and degradation of the c-Myc oncoprotein. This inhibition impairs downstream oncogenic signaling pathways and effectively suppresses cell proliferation and tumor growth in basal-like breast cancer models.
