Biological Activity

Thalidomide-O-COOH (Cereblon ligand 3) is a functionalized thalidomide derivative that acts as a ligand for the Cereblon (CRBN) E3 ubiquitin ligase. It is widely utilized as a building block in the synthesis of proteolysis targeting chimeras (PROTACs). By tethering to a target protein-binding moiety via a flexible linker, this agent recruits the CRBN E3 ligase complex to promote selective target protein ubiquitination and subsequent proteasomal degradation. FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
