Biological Activity

TH287 is a potent and selective MTH1 (NUDT1) inhibitor with an IC50 of. By suppressing MTH1, it impairs the sanitization of oxidized nucleotide pools, leading to the incorporation of damaged nucleotides such as 8-oxo-dGTP into genomic DNA. This accumulation induces double-strand DNA breaks, activates DNA damage response pathways, and ultimately triggers selective cytotoxicity in cancer cells. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
