TH1020 is a potent and selective antagonist of the Toll-like receptor 5 (TLR5)/flagellin complex that exhibits selectivity over TLR2, TLR3, TLR4, TLR7, and TLR8. By disrupting the functional interaction between flagellin and TLR5, TH1020 effectively blocks downstream NF-kB signaling cascades and downstream inflammatory responses, suppressing the activation of innate immune defense mechanisms.
