Evofosfamide (TH-302) is a selective hypoxia-activated prodrug that targets hypoxic tumor microenvironments. Under hypoxic conditions, TH-302 is enzymatically reduced to release the DNA-alkylating agent bromo-isophosphoramide mustard. This conversion leads to DNA cross-linking, down-regulation of cyclin proteins, cell-cycle arrest, and induction of apoptosis. Consequently, TH-302 exhibits potent, dose-dependent cytotoxicity selectively under hypoxia across various cancer cell lines. [1]

- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
