Tetramethylcurcumin (FLLL31) is a curcumin-derived small-molecule STAT3 inhibitor that selectively binds Janus kinase 2 and the STAT3 Src homology-2 domain. By targeting the JAK2/STAT3 pathway, it inhibits STAT3 phosphorylation, DNA-binding activity, and gene transactivation. These molecular effects impede downstream oncogenic signaling, leading to reduced cell invasion, suppressed colony formation, and the induction of apoptosis in breast and pancreatic cancer cell lines. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
