Tetracaine HCl is a potent local anesthetic and an allosteric inhibitor of channel function, primarily targeting ryanodine receptors and voltage-gated sodium channels. By inhibiting calcium release channels in the sarcoplasmic reticulum, tetracaine alters intracellular calcium signaling and suppresses spontaneous calcium release events. High concentrations of the compound lead to a complete blockade of calcium release, thereby modulating excitation-contraction coupling in cardiac and skeletal muscle cells.

- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
