Biological Activity

Teriflunomide (A77 1726) is an immunomodulatory agent and a potent inhibitor of dihydroorotate dehydrogenase (DHODH), the key enzyme in de novo pyrimidine synthesis. By blocking DHODH activity in mitochondria, it depletes intracellular pyrimidine nucleotide pools, suppressing DNA and RNA synthesis in rapidly proliferating cells. This action leads to G1 cell cycle arrest and inhibition of activated T and B lymphocyte proliferation without inducing cell death. [1][2]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
