Tenovin-1 is a small-molecule p53 activator and inhibitor of the sirtuin family protein deacetylases SirT1 and SirT2, as well as dihydroorotate dehydrogenase (DHODH). By inhibiting sirtuin-mediated deacetylase activity, Tenovin-1 protects p53 from MDM2-mediated ubiquitination and proteasomal degradation. This leads to the accumulation and activation of functional p53, triggering downstream cell cycle arrest and apoptosis in p53-expressing cancer cells. [1]
