Biological Activity

Tenofovir Alafenamide is an orally bioavailable prodrug of tenofovir that functions as a viral reverse transcriptase inhibitor. Upon intracellular uptake, it is cleaved primarily by cathepsin A to release tenofovir, which is subsequently phosphorylated into its active metabolite, tenofovir diphosphate. This active form competes with endogenous deoxyadenosine triphosphate for incorporation into viral DNA, causing DNA chain termination and suppressing replication of HIV-1 and hepatitis B virus. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
