Biological Activity

Tazemetostat hydrobromide (EPZ-6438 hydrobromide) is a potent, selective, orally available EZH2 inhibitor that inhibits EZH2 with IC50 values ofin peptide assays andin nucleosome assays, while inhibiting EZH1 with an IC50 of. By blocking EZH2 catalytic activity, it decreases histone H3 lysine 27 trimethylation (H3K27me3), suppressing polycomb repressive complex 2 (PRC2)-mediated transcriptional silencing and inducing anti-proliferative phenotypes in target cells. FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
