Talazoparib (BMN-673) is a potent inhibitor of PARP1 (IC50 0.57 nM) and PARP2. By selectively binding to PARP enzymes and trapping PARP-DNA complexes, it blocks single-strand DNA break repair and disrupts base excision repair pathways. This trapping leads to persistent double-strand breaks during replication, driving synthetic lethality and cell death in homologous recombination-deficient tumors. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
