Biological Activity

TAK-659 Hydrochloride is a potent inhibitor of spleen tyrosine kinase (SYK) with an IC50 of 3.2 nM, while also displaying potent activity against FLT3. By blocking SYK signaling within the B-cell receptor pathway, TAK-659 disrupts downstream AKT/mTOR and ERK activation cascades. This pathway suppression leads to inhibited cellular proliferation, microenvironment signal disruption, and increased apoptosis in hematologic malignancy models.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
