Tacrolimus monohydrate (FK506) is a macrolide antibiotic and potent calcineurin phosphatase inhibitor that acts by binding to FK506-binding protein (FKBP). The resulting FKBP-FK506 complex inhibits calcineurin activity, thereby blocking calcium-dependent signaling pathways, including interleukin-2 gene transcription, nitric oxide synthase activation, cell degranulation, and apoptosis. Consequently, this compound exhibits marked immunosuppressive, neuroregenerative, and neuroprotective properties. [1][2]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
