SU9516 is a 3-substituted indolinone cyclin-dependent kinase (CDK) inhibitor targeting CDK2, CDK1, and CDK4. By inhibiting CDK complexes, SU9516 blocks cell cycle progression, leading to G1/S and G2/M phase cell cycle arrest. Furthermore, inhibition of CDK activity suppresses downstream RB phosphorylation and downregulates anti-apoptotic proteins such as MCL1, ultimately triggering mitochondrial dysfunction and promoting apoptotic cell death in cancer cells.
