Biological Activity

Smurf1-IN-A01 is a small-molecule inhibitor targeting the E3 ubiquitin ligase Smurf1. By selectively inhibiting Smurf1 enzymatic activity, this compound prevents Smurf1-mediated ubiquitin-proteasome degradation of downstream Smad1 and Smad5 proteins. This suppression preserves intracellular Smad levels and key signaling components, thereby accelerating cellular BMP-2 signal responsiveness and enhancing overall bone morphogenetic protein pathway activation. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
