SMER28 is a small-molecule enhancer of autophagy that induces autophagic flux independently of the rapamycin/mTOR signaling pathway in mammalian cells. By stimulating autophagosome formation, SMER28 accelerates the clearance of toxic intracellular protein aggregates, such as mutant huntingtin and alpha-synuclein. This rapamycin-independent stimulation of autophagy reduces protein-misfolding neurotoxicity and confers cytoprotective effects in cell models of neurodegenerative conditions. [1]

- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
