Biological Activity

SJ-172550 is a small-molecule inhibitor of MDMX that reversibly binds to the p53-binding pocket of MDMX. By disrupting the MDMX-p53 interaction, it restores p53 pathway activity and promotes downstream p53-mediated transcriptional signaling. Consequently, SJ-172550 effectively kills retinoblastoma cells with amplified MDMX expression, and its cytotoxic effects are additive when combined with MDM2 inhibitors. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
