Biological Activity

Siramesine (Lu 28-179) is a potent, selective agonist of the σ2 receptor compared to the σ1 receptor subtype. By binding to σ2 receptors, siramesine triggers lysosomal membrane permeabilization and elevates intracellular reactive oxygen species levels, causing lysosomal dysfunction and rapid destabilization. This cascade disrupts cellular homeostasis, promoting caspase-independent cell death across various cancer cell lines. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
