Biological Activity

(Rac)-Shikonin is a potent and specific inhibitor of pyruvate kinase M2 (PKM2) that also inhibits TMEM16A chloride channel activity. By targeting PKM2, it disrupts aerobic glycolysis in tumor cells. Furthermore, (Rac)-Shikonin exerts significant anti-inflammatory effects by suppressing tumor necrosis factor-α (TNF-α) production and inhibiting the NF-κB signaling pathway through targeted proteasome inhibition, ultimately suppressing cellular proliferation and inflammatory responses.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
