SH-4-54 is a potent STAT inhibitor that targets STAT3 (Kd of 300 nM) and STAT5 (Kd of. By blocking STAT3 and STAT5 phosphorylation and transcriptional activity, SH-4-54 suppresses downstream signal transduction through STAT pathways. This inhibition reduces expression of key downstream targets, leading to impaired cell proliferation, suppression of stemness markers, and induction of apoptosis in brain tumor stem cells and glioblastoma models. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
