Biological Activity

SCR7 is a selective DNA ligase IV inhibitor that blocks non-homologous end joining (NHEJ), a key pathway in DNA double-strand break repair. By suppressing NHEJ activity, SCR7 shunts repair pathways toward homology-directed repair (HDR), thereby enhancing HDR-mediated CRISPR/Cas9 genome editing efficiency in mammalian cells and mouse embryos. This selective inhibition promotes precise gene modifications while suppressing error-prone end joining. [1][2]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
