Biological Activity

SAR407899 hydrochloride is an ATP-competitive Rho kinase (ROCK) inhibitor, exhibiting inhibition constant (Ki) values of 36 nM for human ROCK-2 and 41 nM for rat ROCK-2. By suppressing ROCK-mediated phosphorylation of myosin phosphatase, it inhibits thrombin-induced stress fiber formation, platelet-derived growth factor-induced cellular proliferation, and monocyte chemotactic protein-1-stimulated chemotaxis, ultimately resulting in concentration-dependent relaxation of precontracted vascular smooth muscle tissue.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
