Safusidenib is an orally bioavailable, selective mutant IDH1 inhibitor that targets IDH1R132H (IC50 = 15 nM) and IDH1R132C (IC50 = 130 nM) while showing minimal activity against the wild-type enzyme. Inhibition of mutant IDH1 suppresses the aberrant production of 2-hydroxyglutarate, restoring normal cellular differentiation and exhibiting marked antitumor activity in preclinical glioma and chondrosarcoma models. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
