Biological Activity

S-Ruxolitinib (INCB018424) is a potent and selective inhibitor of JAK1 (IC50 3.3 nM) and JAK2 (IC50 2.8 nM), displaying over 130-fold selectivity for JAK1 and JAK2 versus JAK3. By blocking JAK1 and JAK2, the compound disrupts the JAK/STAT signaling pathway and downregulates STAT3 phosphorylation. This inhibition suppresses inflammatory cytokine production, reduces cellular proliferation, and induces apoptosis in JAK-dependent hematopoietic neoplastic cells. [1]

A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
